Fungi from the order Cephalothecales (Sordariomycetes, Ascomycota) stands as one of the elusive groups where substantial gaps persist in understanding of various facets of their life. Although many members are wood-inhabiting or soil-born fungi, some induce infections in humans and animals. The challenges posed by Cephalothecales include issues with ambiguous taxonomy, absence of clearly defined generic and species concepts, polyphyletic nature of asexual morphs, the lack of genome and DNA sequences derived from reference strains, and the inability to confidently identify strains obtained from clinical samples, resulting in potentially incorrect treatment. Integration of various biosystematic approaches will ultimately result in understanding their evolution and a rational, phylogeny-based classification with systematic, ecological, and economic implications. The use of standardized EUCAST protocols will elucidate their sensitivities towards antifungal agents. We will delve into realm of production of biological compounds and asses their bioactivities.
